Abstract:

With the specific aim of developing a practical methodology for producing a spectrum of leukocyte typing reagents, two methods were applied to cytotoxic antisera developed in the rhesus monkey by immunization with allogeneic leukocytes a absorption analysis a methodical absorption of aliquots of antiserum by all positive reacting cells of a small test panel of leukocyte donors and b selective absorption absorption of an antiserum with leukocytes pooled from donors selected on the basis of medium reactivity of their leukocytes with the unabsorbed serum. Although the majority of the 12 reagents that were developed were unquestionably polyspecific, the highly selective quality of the reagents permitted an antigenic profiling of the monkey population so that it was possible to gain a general impression of both the number of leukocyte factors shared by any two individuals and the number by which they differed.
