Abstract:

The Final Report covers work performed during the period August, 1977 to June, 1979. The work presented in this report deals primarily with the physical characterization and attempted separation of TNT Reductase and NADH Oxidase activities. The data accumulated using isoelectric focusing, cellulose-based ion exchange chromatography and polyacrylamide gel electrophoresis in conjunction with denaturation and chemical modification techniques appear to support the hypothesis that with TNT Reductase and NADH Oxidase activities reside on a single macromolecule complex and that the two activities may be inseparable. Oxygen elimination experiments as well as radioisotope trace studies indicate that molecular oxygen is not the electron acceptor in the NADH Oxidase reaction. Dithioerythritol DTE does not appear to be related to the cause of the Luciferase glow effect. Author
