Functional genetics for predisposition to development of Type 2 Diabetes in obese individuals

Obesity is a major risk factor for Type 2 Diabetes T2D yet the majority of obese subjects never develop T2D and their metabolic tissues never lose insulin sensitivity. This suggests the key role of genetic predisposition to T2D development in obesity. This proposal assesses for the first time whether single nucleotide variations SNVs within three genes, PIKFYVE, FIG4 and VAC14, encoding proteins that assemble in a single complex to regulate glucose homeostasis, are associated with T2D in obesity. We performed exome sequencing in abdominal fat derived from bariaric surgeries of 96 morbidly obese individuals with or without T2D 48 subjectsgroup.
