Abstract:

Research apparatus was constructed to include optical systems, high-intensity, light sources, and automatic programming and recording circuits. Histological-biochemical techniques for assay of light exposed retinae, behavioral techniques for establishing and maintaining behavoir in monkeys with which to map their luminosity functions, and electrophysiological techniques to record and monitor ERGs following exposure to light of varying intensities was developed and refined. Eight enzymes were found to show consistent differences in their absorption of nitroblue tetrazolium salt when the staining was done in dark adapted versus light adapted retinae. Refinements of staining, sectioning, and freezing techniques should produce a method for detecting retinal changes produced by light energy below burn threshold. ERG procedures have been refined for use in monkeys. Behavioral techniques have been used to train four monkeys to respond differentially to two stimulus conditions, one representing an abovethreshold stimulus and the other a below-threshold stimulus.
