Plant extracts have been surveyed in an effort to locate natural principles capable of mediating potent, specific inhibition of malaria DNA polymerase alpha. Several extracts containing putative polymerase inhibitors have been shown to inhibit cultured Plasmodium falciparum. Also underway are efforts to clone and express the gene for P. falciparum DNA polymerases alpha and delta, which would provide access to unlimited amounts of the enzyme far screening and isolation of polymerase alpha and delta inhibitors. In addition we have developed a strategy that should permit the chemical synthesis of the gene for polymerase delta the nucleotide sequence of which has been reported and plan to synthesize and express the entire gene. Malarial, Plasmodium falciparum, DNA polymerase, natural products, plants.
