Abstract:
We are testing the hypothesis that the balance between excitation and inhibition is skewed in TSC brains and that this synaptic dysregulation is caused by altered protein synthesis. We have found that the imbalance between excitation and inhibition using biochemical assays and electrophysiology. Furthermore, calcium imaging show hyperexcitable neuronal activities in TSC-1 deleted neurons. These results further support our hypothesis. Correction of this synaptic dysregulation will hopefully provide a new therapeutic target to neurological symptoms in TSC such as epilepsy, mental retardation and autistic behaviors.
