Abstract:

The goal of these studies is to monitor changes occurring in host macrophages using a cell culture model of the early establishment stage of anthrax. Global changes in cultured murine macrophage expression of mRNA and protein, in response to B. anthracis and anthrax lethal toxin challenges, were assessed for 8 hours via microarray and MudPIT technologies and compared to mock-challenged cells. While most host genesproteins did not change expression levels in response to the challenge, there were specific classes of both up-regulated and down-regulated expression patterns that did. These 2 broad classes were divided into two distinct subclasses responses common to many types of bacterial challenges and those that appeared to be B. anthracis-specific. The later category holds promise of targets for early diagnosis and perhaps intervention points.
