The object of this proposal is to understand the tumor suppressor function of maspin in prostate, and to explore maspins role in normal prostate development. Maspin knockout mouse model will be employed to study the effects of loss of maspin function on mouse prostate tumorigenesis and development. We hypothesize that the presence of maspin by stable transfection or by adenovirus mediated maspin gene delivery will prevent or delay prostatic tumorigenesis and metastasis, while loss of maspin in mouse model will render it more susceptible to tumor formation and metastasis.
