Abstract:

The hypothesis that secreted heat shock protein gp96 will trigger an immune response was tested in a tumor system in experimental animals. It was found that gp96 is able to induce tumor specific CD8 CTL without the help of CD4 cells. The postulated molecular and cellular mechanism involves the following steps 1 gp96-Ig peptide secretion 2 Binding to endocytosis receptor on dendritic cells 3 Dendritic cell activationmaturation, including up-regulated expression of CD8O86 4 Uptake of gp96 and routing of peptides to endoplasmic reticulum 5 loading of trimmed peptides on class I MHC molecules and transport to the cell surface 6 CD8- pre-CTL activation independent of CD4 T cell help.
