Abstract:

In Year 1 of this grant, we are establishing procedures for determining the ability of the phosphonic acid diphenylureas to inhibit PC3 prostate carcinoma cells in the nude mouse xenograft. The initial experiments have not confirmed that the phosphonic acid diphenylureas inhibit prostate cancer in the nude mouse xenograft. Additional experiments are necessary to determine the reason for the differences in results in the in vitro and in vivo experiments. We postulate that the i.p. injection of the treatments into the peritoneal cavity does not deliver the phosphonic acid diphenylureas to the tumor. In other experiments, the phosphonic acid diphenylureas have been shown to be potent inhibitors of metalloproteinase MMP activity in prostate carcinoma cells. We postulate that these compounds inhibit MMP secretion or synthesis. Additional experiments are proposed to test this hypothesis.
