Abstract:
The effects of retinoic acid RA in breast cancer have been widely studied. Treatment with RA leads to either growth arrest or apoptosis in breast cancer cells. The cyclin-dependent kinase inhibitor p2l WAF1CIP1 may be involved in terminal differentiation associated growth arrest. We showed in this study that PMLRAR alpha increased the transcription of p21WAF1CIP1 gene and the activation was further induced by RA treatment. Deletion analysis revealed a region upstream of the p2lWAFJClPl promoter that is required for transactivation by PMLRAR alpha. Transient transfection of PMLRAR alpha in cells increased the endogenous p2l WAF1CIPl protein levels.
