Abstract:

The goal of this project is to determine the role of the Src Homology 2 SH2 domain protein, Grb7, in tyrosine kinase signal transduction. Grb7 is overexpressed in approximately 20 of breast cancers in combination with the HER2 receptor tyrosine kinase. Our initial goals of this project were to determine the role of Grb7 overexpression on breast cancer cell behavior as well as to identify proteins that interact with Grb7. Our initial studies have not indicated a specific phenotype induced by Grb7 overexpression or specific proteins that can associate with Grb7. In order to determine a function for Grb7, we are currently generating a mouse deficient in Grb7.
