Abstract:

The goal of this project is to elucidate the mechanism of estrogen receptor ER action through the identification of Proteins involved in ER signaling using a genetic approach. We have utilized the phenotype of a mutant ER G4O0V ER, deficient in its ability to bind hormone, as the basis for a dosage suppression screen in yeast. Screening a high-copy yeast genomic library, we have isolated a gene that dramatically suppresses the phenotype of G4OOV ER by increasing its ability to bind hormone. Overexpression of this protein also increases the activity of both wild type estrogen and glucocorticoid receptors GR. The suppressor is the yeast homologue of the human p23 protein, a component of the molecular chaperonin complex bound to many unliganded steroid receptors.
