In the time period covered by this report we have carried out three sets of preliminary studies. Pulsed field gel electrophoresis has been used to examine possible amplified regions on chromosome 20 implicated in breast cancer to provide target areas of the genome for further study. Various methods for producing dense arrays have been developed and evaluated. Elaborations on these methods have been designed that promise to produce high quality cDNA profiling. A new method for profiling DNA changes has been developed that uses different genomic gel fragments in a single gel lane. This method appears to be extremely promising and its use in the search for DNA changes important in breast cancer will be a major focus of future studies.
