Thirteen dengue virus-specific, cytotoxic CD4 CD8- T cell clones were established from a donor who was infected with dengue-3 virus. Six patterns of virus specificities were determined. Two serotype-specific clones recognized only dengue-3 virus. Two dengue subcomplex-specific clones recognized dengue-2, -3 and -4 virus, and one subcomplex-specific clone recognized dengue-1, -2, and -3 virus. Four dengue serotype-crossreactive clones recognized dengue-1, -2, -3 and -4 virus. One flavivirus-crossreactive clone recognized dengue-1, -2, -3, and -4 virus and West Nile virus WNV, but did not recognize yellow fever virus YFV, while three flavivirus-crossreactive clones recognized dengue-1, -2, -3 and -4 virus, WNV and YFV. We also examined the recognition of purified NS3 proteins of degue-3 virus and WNV by these T cell clones. One serotype-specific clone, two dengue subcomplexspecific clones and three dengue serotype- crossreactive clones recognized NS3 of dengue-3 virus. One flavivirus- crossreactive clone recognized NS3 of dengue-3 virus and WNV.
