Abstract:

The goal of this contract was to isolate antigens of Leishmania major and test them for their ability to impart protection against parasite challenge in mice. We prepared monoclonal antibody producing hybridomas from splenocytes of mice immunized with fixed promastigotes. We induced ascites production in mice hybridomas ip, and purified antibody from ascites. The antigens recognized by selected monoclonal antibodies were assessed by immunoprecipitation and Western blot analysis. In addition, we developed a radioimmunoassay for detecting parasite antigens expressed on the surface of infected cells. We used this assay to screen monoclonal antibodies that recognized expressed strains. Keywords Leishmaniasis Macrophages Antileishmanial drugs.
