Abstract:
The objective of this study was to determine the feasibility of identifying short-term in vivo or in vitro toxicity tests which could be included in a battery of such tests as part of a toxicological screening program. The study was performed by a panel of ten toxicology experts and a contractor management team, which included scientific information specialists. Six compounds benzene, cadmium, formaldehyde, phosphorus, phosgene, oxides of nitrogen were chosen as models for analysis by a matrix format. Each matrix was supported by a review article. Panel members selected predictive endpoints from acute and subchronic data taken from the literature. The panel also recommended short-term tests relevant to the endpoints for each compound. Position papers were developed for pharmacokinetics behavioral toxicity in vitro testing reproductive assessment testing and a concept for toxicological testing. Bibliographies were prepared for the matrix reviews, each position paper, and one for the overall study. Based on the analyses of the matrices, the position papers and their collective experience, the panel developed recommendations for short-term tests for a minimal toxicology screening program and pointed out gaps wherein additional research was required.
