Abstract:
A program on Synthesis of Cocaine Metabolites and Ring-Radiolabeled Cocaine has succeeded in producing the four required metabolites. Progress has been made on the development of a synthetic route to cocaine which would permit the preparation of that compound with a 14C label located in the ring at metabolically stable positions. However obstacles have been encountered in the synthesis which have prevented its completion. In brief, the authors have been unable to prepare the important intermediate succindialdehyde or a suitable derivative in a sufficiently high yield by a route that would allow for the introduction of 14C and subsequently undergo reaction to form the necessary intermediate tropinone derivative. This report describes attempts to circumvent these difficulties by either a isolating the succindialdehyde after generating it in the condensation inhibiting environment, b oxidizing 1,4-butanediol by a different method and thus avoiding the problem of inhibition, or c introducing the label through some precursor other than succinic acid.
