The objective of this proposal is to evaluate whether the whole-genome LOF screen in vivo is feasible by exploiting a multiplexed AsCpf1 based screen strategy, and thus can be used to systematically discover effective therapy targets in tumor in vivo setting. The scope of the research is to demonstrate and validate this novel function screen approach as an in vivo orthotopoic discovery platform to identify novel immunotherapeutic targets towards more effective anti-PDAC therapy.
