The project aims to develop a novel technology to preserve vascular composite allografts for extended periods. This project uses a rat limb transplant model and aims to preserve grafts for 3 days or more. In this first year, we developed a machine perfusion protocol for rat limbs. Several aspects had to be optimized, including media, additives, oxygen carriers among others to identify a protocol that does not lead to ischemia, exhibits sustained low vascular resistance, and energy levels. We further started evaluation of the perfused grafts by transplantation with success in the first tests. We also started optimization of the supercooled graft preservation, which will be the main goal in year 2.
