Abstract:

Islets are composed of or 5 endocrine cell types that perform complementary functions to maintain proper glucose homeostasis. This cellular heterogeneity impedes our ability to understand the precise transcriptional repertoire and regulatory landscape of each cell type and to determine how these programs in each cell type are perturbed in type 2 diabetes T2D. The overarching goal of this project is to determine, with single cell resolution, changes in cellular composition and cell-specific gene expression programs elicited by T2D in human islets using innovative single cell transcriptomic scRNA-seq Aim 1 and epigenomic scATAC-seq Aim 2 technologies.
