This project focuses on addressing cartilage lubricant failure in post-traumatic osteoarthritis PTOA via intra-articular lubricinproteoglycan 4 PRG4 gene therapy. Our findings thus far indicate that a single intra-articular injection of adeno-associated virus AAV bearing the PRG4 gene in a rabbit ACL transection ACLT model resulted intransgene expression until the end-point of the experiment at 8 weeks post-op, and that the injection was significantly chondroprotective, while injections of control virus AAVGFP were not. These data indicate that PRG4 gene therapy is a viable option for slowing the progression of PTOA, a finding that warrants progression to a large animal model.
