Abstract:

The studies elucidate the mechanism of action of chloroquine at the molecular level. The results of the study are relevant to the practical problem of drug resistance in malaria in that they lead to the following conclusions 1 It is doubtful that further development of intercalative compounds will yield superior drugs for chloroquine resistant malaria. The selective toxicity for plasmodia of such substances as a group will be subject to the same limitations as that of chloroquine or quinacrine. 2 Barring radically new developments, the most promising approach to treating chloroquine resistant falciparum malaria will be the administration of synergistic mixtures of existing drugs which potentiate each others antiplasmodial actions at pharmacologically tolerated levels of the individual substances.
