The technique of potential-step chronoabsorptometry has been evaluated as applied to the measurement of homogeneous electron transfer reactions. In particular, the reaction of horse heart cytochrome c with methyl viologen radical cation was examined as an example of biological electron transfer using a non-physiological reductant. Theoretical and experimental considerations affecting the measurement of rate constants are discussed. These include the deconvolution of time-dependent spectra and the influence of concentration and diffusion coefficient ratios.
