Protein Components of Human Erythrocyte Membrane.

Two protein components, PI and PII, have been isolated from the membrane of erythrocytes of fresh blood. PI was extracted with a low ionic strength, and PII with a high ionic medium. After purification through Sephadex G-200 both gave characteristic protein spectra with maximum absorbency at 277-279 millimicron. When subjected to polyacrylamide gel electrophoresis, PI and PII were resolved in several polypeptide chains which is indicative of mixtures of heterogeneous proteins. PI and PII represent 50 of the total membrane proteins and contain very small quantities of lipid, carbohydrate and sialic acid. Amino acid analysis and molecular weight determination show that PI might be the structural membrane protein spectrin isolated by other investigators. Author
