A method was developed for the isolation of urinary testosterone from urinary testosterone glucuronide, followed by purification of the testosterone on an LH-20 column, and quantitation by radioimmunoassay. Reasonable testosterone excretion values, in urine samples from normal volunteers, compatible with published data, were obtained. Methods for the isolation of testosterone, androsteneidione, and 11 beta-hydroxyandrostenedione from plasma have been studied, tested and evaluated. Satisfactory isolations of the steroids of interest were obtained using LH-20 columns. A method for preparing plasma extracts containing the steroids of interest, which were adequate from the quantitative point of view, and could be utilized satisfactorily on LH-20 columns was developed. It is felt that the extraction and isolation procedure is qualitatively and quantitatively satisfactory, and suitable for routine use on multiple columns run simultaneously. Author Modified Abstract
