Automated Quantitation of A and B Blood Group Substances.

An automated assay for the detection and quantitation of soluble blood group antigens is described. Variables such as reaction medium, type and amount of antibody employed, and reaction temperature were investigated and optimal conditions for increased sensitivity, specifity, and precision were determined. This AutoAnalyzer system utilizing modified hemagglutination manifolds and standardized with an NIH 500 microgramml standard has the ability to detect andor quantitate blood group substance A and B at the microgram level. Limitations of the assay are discussed. Rationale behind the need for screening parenteral fluids, especially in the military population is advanced. Author
