Antimalarial activity of test compounds supplied by the Division of Medicinal Chemistry of the Walter Reed Army Institute of Research were evaluated in a Plasmodium knowlesi-rhesus monkey test system. During the 5-year testing period, 272 antimalarial tests of 35 days duration were conducted using 111 different compounds or combinations of compounds. A toxicity test to determine a maximum tolerated drug dose was performed for many test drugs, preceding their use in antimalarial tests. The report is a summation of results with emphasis on the presence or absence of malaria deaths and the duration of parasitemia in test groups of monkeys compared with both untreated control monkeys and reference drug control monkeys which received a curative dose of pyrimethamine.
