Chromosomal changes of Yoshida ascites sarcoma cells induced in vitro by the minimal effective concentration MEC of 4-nitroquinoline-1-oxide 4NQO were studied. Yoshida sarcoma cells were taken from Donryu rats bearing 4-day-old tumor ascites. Culture medium was Eagles MEM 79.9 supplemented with calf serum 20 and Bactopeptone 0.1. The cell number of inoculum was 50,000ml. The concentration 10 to the minus 8th power M of 4NQO was determined as the MEC of this substance for Yoshida sarcoma cells in vitro after repeated cell counts. The predominant chromosomal changes of cells cultivated in the medium containing the MEC of 4NQO were increased chromatid break and chromatid exchange. These changes were observed during 30 hours after the cultivation. During the period, persistent nucleoli in late prophase, prometaphase and metaphase were detected. They showed changed stainability. It is possible that both the chromosomal and nucleolar abnormalities are the products of 4NQO-inhibited DNA and RNA synthesis. Author
