Abstract:

The colony-forming unit CFU is thought to be a the hematopoietic stem cell, and CFU methodology has been used extensively to evaluate radiation damage to the hematopoietic system in mice. Although the method works satisfactorily, the counting of macroscopic spleen nodules is laborious, time-consuming, and, to a certain extent, subjective. A staining-clearing method has been developed which greatly facilitates and renders this method less tedious and more reproducible. The spleen nodules appear as blue-green spheres in a nearly colorless spleen parenchyma. Estimates of radiation sensitivity Do of CFU are not altered by the use of the staining-clearing procedure. However, estimates of the CFU count nucleated marrow cells, or CFU contentfemur, are reduced when the staining technique is used. Author
