Abstract:
The isoelectric point of Cl. perfringens anatoxin, purified via precipitation with HCl at pH 3.8-4.0, comprised 4.8-5.0 upon determination on the MMED apparatus multimembranous electric decantator. Upon optimum conditions of anatoxin purification by the method of electrophoretic convection on the MMED apparatus, a purification coefficient was obtained equaling 1.6-2.1, at 50 preparation yield, and the activity of 10-100 BU per one milligram of protein nitrogen. When ECD-1 electric convection device was used, a preparation was obtained with purification coefficient of 2.7-3.6, up to 36 yield, and 7-30 BUmg of protein nitrogen activity. With four-stage purification of anatoxin Cl. perfringens acid alcohol 2-stage purification with the electroconvection method, a highly purified preparation of Cl. perfringens anatoxin was obtained, with activity up to 1340-1600 BUmg of protein nitrogen. The method of electrophoretic convection with the use of MMED and ECD-1 apparatus can be employed for additional purification of Cl. perfringens anatoxins, obtained as a result of one-state purification precipitation with HCl, at pH 3.8-5.0, or two-stage purification precipitation with HCl with subsequent precipitation with alcohol.
