Abstract:

Rapid prophylactic digitalization of a normal heart may be a helpful pharmacologic device in elucidating the mechanism of action of other cardiotropic chemical substances. The objective of this study was to determine whether the effects of acetyl strophanthidin AS can be assessed under such conditions. It was concluded that the ED50 of AS for producing ventricular arrhythmias in normal dogs is 54.5 microgramskgiv. Doses up to and including those that cause ventricular tachycardia do not produce either atrial arrhythmias or significant conduction disturbances. Although the classical digitalis-induced rate and EKG waveform changes may appear after administration of AS, they have no value in predicting the subsequent development of ventricular arrhythmias. As the administered dose of AS is increased, there are no signs produced in the dog which precede and warn of the toxic dose. Author
