Cobrotoxin contains six disulfide bonds and is devoid of sulfhydryl groups. On reduction with beta-mercaptoethanol, cobrotoxin displays 11.6 to 12.0 sulfhydryl groups and loses the lethality concurrently. The inert, fully reduced cobrotoxin yields biologically active product with complete lethality and cross-reaction with antisera to cobrotoxin, specific rotation close to the native value, and an infrared spectrum identical to native cobrotoxin upon reoxidation. The results of the study clearly indicate that the integrity of the disulfide bonds in cobrotoxin is essential for lethality. Author
