Abstract:
Brain slices from the rat were allowed to respire for 2 hr in a modified Tyrodes solution containing the monoamine oxidase inhibitor 1-phenyl-2-hydrazinopropane alpha-methyl phenyl hydrazine, JB-516. The filtered Tyrodes solution was shown to contain 5-hydroxytryptamine Serotonin, 5-HT by both bioassay and fluorometric techniques. Preliminary evidence indicates that the amount of 5-HT in the filtrate is increased significantly by adding reserpine and JB-516 to the incubation medium. These findings suggest that a sensitive bioassay procedure, coupled with the use of brain slices. offers an approach for the study of factors affecting the storage and release of endogenous 5-HT.
