Abstract:
As part of a study for the elucidation of the structure of saxitoxin, the synthesis was undertaken of a possible C8 fragment resulting from the degradation of the natural product. The preparation of 7-methyl-2-oxo2,4,5,6- tetrahydropyrrolo 1,2-c-pyrimidine, a new ring system, is described. The selective oxidation to the desired 7-methyl-2-oxo-2,4,5,6-tetrahydropyrrolo1,2- c pyrimidine was accomplished with a dilute neutral aqueous solution of potassium permanganate at room temperature overnight. The physical and analytical properties of 7-methyl-2-oxo-2,4,5,6-tetrahydropyrrolo1,2-c pyrimidine were not identical with those of the product obtained from the natural product. The 8-methyl derivative is the correct structure of one of the degradation products of saxitoxin.
