A new bioluminescent chamber was designed and constructed and has resulted in a three-fold increase in reaction sensitivity. For use in conjunction with the new chamber, a Photovolt 520 photometer and a Photovolt 44 linear recorder were purchased. These instruments offer the advantage of greater sensitivity, increased stabil ity, greater portability, and a decrease in maintenance costs. A Branson Ultrasonic Oscillator has been purchased and is now being used for the disruption of microorganisms. With this instrument, ATP has been detected in Serratia marescens with a fivefold increase in sensitivity over that found previously. After measuring ATP response with varying concentrations of enzyme, it was found that the maximum response for concentrations of ATP was obtained by using 20 mg of lyophilized extractml. Progress was made toward the removal of inherent light light emitted by enzyme extract in the absence of ATP. By the use of ammonium sulfate precipitation of the enzyme, and also by means of calcium phosphate gel adsorption, it was possible to reduce the inherent light to very low levels and still retain a good percentage of the enzymatic activity.
