Direct platelet counts made with the phase microscope on venous blood collected in siliconed test tubes reflected the circulating platelet level. Unavoidable field, pipet, and chamber errors accounted for the 11 single-count errors, which can be reduced by doing multiple counts. Platelet counts on capillary blood from a finger puncture were subject to 24 error and the counts were about 2 12 lower than on venous blood this was probably due to a variable platelet loss in the puncture wound. Other methods of platelet counting are reviewed. In 13 subjects the platelet levels varied greatly between individuals, but the individual platelet counts showed only minor variations during a 5-mo period.
