Abstract : Androgen receptor (AR) plays a critical role in the progression of prostate cancer. This project is directed towards testing the feasibility of targeting AR for lysosomal degradation. We have demonstrated that AR can indeed be targeted for degradation by stimulating the TFEB-lysosome degradation pathway either by increasing the TFEB levels or by activating TFEB using mTORC1 kinase inhibitor, torin 1. Additionally, we determined that the same approach can be used to target the EWS-Fli-1 oncoprotein for degradation.
