Abstract : Lack of a definitive system for the diagnosis and staging of NF1-related tumors is a major obstacle to investigating the molecular basis of tumorigenesis, to our ability to assess prognosis, and consequently, to the rational design and application of stage-specific therapeutic agents (1-3). Locus-specific changes in copy number are a common feature of many types of tumors, including the neurofibromas and malignant peripheral nerve sheath (MPNST) that develop in individuals with NF1 (4-13). Gains or losses at a particular locus are (reviewed in (4) potential biomarkers for the molecular diagnosis and grading of MPNST. Towards the goal of molecular diagnosis and staging of NF1-related tumors, we propose to develop and validate a novel polymerase chain reaction (PCR)-based method, termed QuEST, for rapid and quantitative identification of loci with increases or decreases in genomic copy number in MPNST.
