Abstract : The present grant proposes to look at the effects of blast on the eye of the mouse looking at phenotypic changes in the eye and in the changes in gene expression following a 50psi blast. We are using these data to define biomarkers to predict the severity of the injury and to predict eventual outcomes. We have examined the phenotypic changes in the eye in the BXD strains before and 5 days after a 50psi blast and have observed no strain specific change in either the cornea or the IOP. We have completed the normal retina database containing 222 microarrays from 58 strains of mice. We have also prepared the manuscript describing the database and submitted it to Molecular Vision for publication. The database and annotation are complete and will be released to the public upon the acceptance of the manuscript. We have collected 76 retinas from 19 strains 5 days after a 50psi blast. These retinas are being held for a batch RNA isolation and microarray run. Microarrays were run on an additional 27 strains. Next year we will complete the dataset by adding 84 retinas from 21 BXD strains. We have taken a preliminary examination of the blast dataset and found that Sox l 1 is a relatively good marker for injured retinal ganglion cells. These results are presented in a paper currently under revision for publication in Molecular Vision.
