Abstract : A new very reliable biochemical technique for determining a very small amount (les s than 1 mcg/ml) in the biological fluid was reported. Close correlation was established between the biologically active isoniazid (INH) level of blood plasma and the urinary excretion of free INH determined by our new method in comparison with other biochemical techniques. We were not successful in demonstrating trimodal distribution of the biologically active INH plasma levels in the monkey and in establishing a correlation between blood levels and acetylating activity of monkey liver homogenate. In monkey, the liver is the most active site of acetylation but the kidney and spleen has the capacity too. The electrophoretic study on the concentrated acetone fraction 60 (transfer enzyme) revealed that the pattern of the enzyme for PABA acetylation was quite similar to that for Sulfanilamide, while further investigation seems to be necessary as to a pattern of the transacetylase which is concerned with the acetylation of INH.
