Abstract : Ricin, the highly toxic hemagglutinating protein, was isolated from the seeds of Ricinus Communis L. The defatted castor meal firstly extracted with 10% sodium chloride solution, followed by fractional precipitation with ammonium sulfate, the highly toxic ricin fractions I and II were obtained. Recin I was the most toxic fraction, killing the mouse of 20 Gm. at a dose of 1 mcg. by intraperitoneal injection of 4 days. The qualitative determination of Ricin fractions was by paper chromatographic method. Digestion of Ricin I by pepsin and trypsin was found to take place slowly in 3 days. A prolonged enzymatic digestion was made for 2 weeks, it broke down about 39-48% of low molecular products. Antitoxic serum to Ricin I was prepared by immunization of rabbits with an increasing amount of formalinized toxoid (formalin treated Ricin I). The concentration of 0.5-5% formalin were used resulting about a 100-1000 fold reduction in toxicity of Ricin I. Marked loss of antifenicity occured with formalin at pH 8.5 and above.
